To study the influence of corosolic acid(CRA) on the growth of human lung carcinoma A549 cells and discuss the mechanism of its antitumor action. Methods:MTT method was used to detect the influence of CRA on A549 lung cancer cell growth in vitro under different concentrations. The nude mouse tumor model carrying bioluminescence-marked lung cancer cells A549 was established. IVIS small animal in vivo imaging system was used to detect the growth of solid tumor. The expressions of vascular endothelial growth factor(VEGF) and CD34 in tumor tissues were detected by immunohistochemical analysis. CD34 positive blood vessels were taken as the microvascular density(MVD).
Results:The value of IC50 was 26.8 μg / ml in vitro experiment. CRA of different doses had certain therapeutic effects on A549 solid tumor, the content of VEGF and CD34 proteins also had different degrees of influence.
Conclusion:Banaba leaf extract Corosolic acid has definite therapeutic effects on A549 solid tumor, the mechanism of its antitumor action may be connected with the inhibition of the expression of VEGF and CD34 proteins in the tumor tissue.
